Nedostatak prisutnosti određenog proteina u koži povezan je s aknama, ako je vjerovati novoj studiji objavljenoj u Nature Communications. Prema autorima, kao rezultat istraživanja mogli bi se razviti novi tretmani koji promiču rast ovog novoznačajnog proteina.

Akne su vjerojatno najučestalije bolno stanje kože na svijetu, a procjenjuje se da u nekom trenutku pogađaju oko 650 milijuna ljudi. Ova bolest kože obično se javlja kada se uljne žlijezde začepe, stvarajući mrlje, prištiće i ponekad ciste. Međutim, još uvijek postoje mnoge nepoznanice o nastanku akni.

"Iako su akne jedno od najčešćih dermatoloških stanja, njihova patogeneza ostaje nepotpuno razumljiva", rekla je dr. Christina Philippeos, znanstvena suradnica u Centru za matične stanice i regenerativnu medicinu na King's Collegeu u Londonu.

Kako bi saznali više o aknama, Philippeos i kolege istražili su je li na bilo koji način na njihov nastanak uključen GATA6 (protein koji veže GATA 6), prisutan u folikulu dlake. Bilo je nekoliko nagovještaja da jest.

Prije toga, druga je skupina pokazala da kada se ovaj protein isključi u epidermalnim stanicama koje postavljaju strukturu koja se sastoji od folikula dlake i lojnih žlijezda, lezije će nastati ispunjene materijalom sličnim aknama.

Da bi saznali više, istraživači su pregledali biopsije kože pet zdravih dobrovoljaca i devet pacijenata s različitim stupnjem ozbiljnosti akni. Pregledom su otkrili da zdravi ljudi imaju visoku razinu GATA6, dok je u slučajevima akni razina GATA6 bila smanjena.

Istraživači su razvili ljudski lojni organoidni model koji je na kraju pokazao da je GATA6 uključen u nekoliko fizioloških procesa koji reguliraju gornju pilosebacealnu jedinicu (PSU). Jedan takav postupak povezan je s proizvodnjom i diferencijacijom keratinocita, koji su primarne, najčešće stanice pronađene u najudaljenijem sloju kože.

"Akne su bolest specifična za ljude i niti jedan životinjski model u potpunosti ne rekapitulira svoje patološke značajke. Stoga su se istraživanja akni u velikoj mjeri oslanjala na translacijske histopatološke studije i studije zasnovane na stanicama. Može biti popriličan izazov pokazati da su nalazi viđeni in vitro reprezentativni za bolest kod ljudi. Štoviše, pristup histološkim uzorcima kože s aknama težak je, jer je za to potrebna biopsija pacijenta, koja se obično izvodi na licu. Ipak, zaista smo uživali u tehničkom izazovu stvaranja malih lojnih 3D organoida i izoliranja folikula dlake od svježe ljudske kože (dobivene od otpada plastične kirurgije) kako bismo stvorili in vitro kulture mini-organa za utišavanje GATA6 u ovim PSU-ima ”, pojasnila je Philippeos za ZME Science.

Štoviše, GATA6 modulira imunološke signale koji doprinose upalnim putovima viđenim kod akni, a također može spriječiti stvaranje začepljenih pora.

No ono što je najvažnije istraživači su otkrili da je ekspresiju GATA6 inducirala retinoična kiselina, proizvod koji se koristi u nekim tretmanima akni.

„Naše istraživanje pokazuje da izotretinoin djelomično djeluje kroz indukciju GATA6. Stoga već postoji tretman temeljen na GATA6! Ali naša je studija još uvijek u fazi otkrivanja. GATA6 nikada prije nije bio uključen u patogenezu akni. Potrebna su daljnja istraživanja kako bi se utvrdilo koji spojevi mogu posebno stimulirati ekspresiju GATA6 kod pacijenata s aknama, prije nego što liječenje krene na probu", rekla je Philippeos.

U budućnosti istraživači planiraju provesti više istraživanja uloge GATA6 u ljudskoj koži. U međuvremenu, Philippeos preporučuje sistemski izotretinoin za liječenje akni, što naziva "trenutnim zlatnim standardom", ali koji također ima više nuspojava.

„Ova studija pokazuje javnosti da je razvoj akni složen, višefaktorski proces. Još uvijek nije u potpunosti razumljivo, ali ovo otkriće baca više svjetla na ove procese, što bi moglo otvoriti nove putove za istraživanje i liječenje ove bolesti ”, zaključuje ona.

Nedostatak prisutnosti određenog proteina u koži povezan je s aknama, ako je vjerovati novoj studiji objavljenoj u Nature Communications. Prema autorima, kao rezultat istraživanja mogli bi se razviti novi tretmani koji promiču rast ovog novoznačajnog proteina.

Akne su vjerojatno najučestalije bolno stanje kože na svijetu, a procjenjuje se da u nekom trenutku pogađaju oko 650 milijuna ljudi. Ova bolest kože obično se javlja kada se uljne žlijezde začepe, stvarajući mrlje, prištiće i ponekad ciste. Međutim, još uvijek postoje mnoge nepoznanice o nastanku akni.

"Iako su akne jedno od najčešćih dermatoloških stanja, njihova patogeneza ostaje nepotpuno razumljiva", rekla je dr. Christina Philippeos, znanstvena suradnica u Centru za matične stanice i regenerativnu medicinu na King's Collegeu u Londonu.

Kako bi saznali više o aknama, Philippeos i kolege istražili su je li na bilo koji način na njihov nastanak uključen GATA6 (protein koji veže GATA 6), prisutan u folikulu dlake. Bilo je nekoliko nagovještaja da jest.

Prije toga, druga je skupina pokazala da kada se ovaj protein isključi u epidermalnim stanicama koje postavljaju strukturu koja se sastoji od folikula dlake i lojnih žlijezda, lezije će nastati ispunjene materijalom sličnim aknama.

Da bi saznali više, istraživači su pregledali biopsije kože pet zdravih dobrovoljaca i devet pacijenata s različitim stupnjem ozbiljnosti akni. Pregledom su otkrili da zdravi ljudi imaju visoku razinu GATA6, dok je u slučajevima akni razina GATA6 bila smanjena.

Istraživači su razvili ljudski lojni organoidni model koji je na kraju pokazao da je GATA6 uključen u nekoliko fizioloških procesa koji reguliraju gornju pilosebacealnu jedinicu (PSU). Jedan takav postupak povezan je s proizvodnjom i diferencijacijom keratinocita, koji su primarne, najčešće stanice pronađene u najudaljenijem sloju kože.

"Akne su bolest specifična za ljude i niti jedan životinjski model u potpunosti ne rekapitulira svoje patološke značajke. Stoga su se istraživanja akni u velikoj mjeri oslanjala na translacijske histopatološke studije i studije zasnovane na stanicama. Može biti popriličan izazov pokazati da su nalazi viđeni in vitro reprezentativni za bolest kod ljudi. Štoviše, pristup histološkim uzorcima kože s aknama težak je, jer je za to potrebna biopsija pacijenta, koja se obično izvodi na licu. Ipak, zaista smo uživali u tehničkom izazovu stvaranja malih lojnih 3D organoida i izoliranja folikula dlake od svježe ljudske kože (dobivene od otpada plastične kirurgije) kako bismo stvorili in vitro kulture mini-organa za utišavanje GATA6 u ovim PSU-ima ”, pojasnila je Philippeos za ZME Science.

Štoviše, GATA6 modulira imunološke signale koji doprinose upalnim putovima viđenim kod akni, a također može spriječiti stvaranje začepljenih pora.

No ono što je najvažnije istraživači su otkrili da je ekspresiju GATA6 inducirala retinoična kiselina, proizvod koji se koristi u nekim tretmanima akni.

„Naše istraživanje pokazuje da izotretinoin djelomično djeluje kroz indukciju GATA6. Stoga već postoji tretman temeljen na GATA6! Ali naša je studija još uvijek u fazi otkrivanja. GATA6 nikada prije nije bio uključen u patogenezu akni. Potrebna su daljnja istraživanja kako bi se utvrdilo koji spojevi mogu posebno stimulirati ekspresiju GATA6 kod pacijenata s aknama, prije nego što liječenje krene na probu", rekla je Philippeos.

U budućnosti istraživači planiraju provesti više istraživanja uloge GATA6 u ljudskoj koži. U međuvremenu, Philippeos preporučuje sistemski izotretinoin za liječenje akni, što naziva "trenutnim zlatnim standardom", ali koji također ima više nuspojava.

„Ova studija pokazuje javnosti da je razvoj akni složen, višefaktorski proces. Još uvijek nije u potpunosti razumljivo, ali ovo otkriće baca više svjetla na ove procese, što bi moglo otvoriti nove putove za istraživanje i liječenje ove bolesti ”, zaključuje ona.

Nedostatak prisutnosti određenog proteina u koži povezan je s aknama, ako je vjerovati novoj studiji objavljenoj u Nature Communications. Prema autorima, kao rezultat istraživanja mogli bi se razviti novi tretmani koji promiču rast ovog novoznačajnog proteina.

Akne su vjerojatno najučestalije bolno stanje kože na svijetu, a procjenjuje se da u nekom trenutku pogađaju oko 650 milijuna ljudi. Ova bolest kože obično se javlja kada se uljne žlijezde začepe, stvarajući mrlje, prištiće i ponekad ciste. Međutim, još uvijek postoje mnoge nepoznanice o nastanku akni.

"Iako su akne jedno od najčešćih dermatoloških stanja, njihova patogeneza ostaje nepotpuno razumljiva", rekla je dr. Christina Philippeos, znanstvena suradnica u Centru za matične stanice i regenerativnu medicinu...
